PPA1 Regulates Systemic Insulin Sensitivity by Maintaining Adipocyte Mitochondria Function as a Novel PPARγ Target Gene

چکیده
Downregulation of mitochondrial function in adipose tissue is considered as one important driver for the development obesity-associated metabolic disorders. Inorganic pyrophosphatase 1 (PPA1) an enzyme that catalyzes hydrolysis inorganic pyrophosphate to phosphate and required anabolism take place cells. Although alteration PPA1 has been related some diseases, importance syndromes never discussed. In this study, we found global knockout mice (PPA1+/–) showed impaired glucose tolerance severe insulin resistance under high-fat-diet feeding. addition, ectopic lipid accumulation were observed. Conversely, overexpression by adeno-associated virus injection can partly reverse disorders PPA1+/– mice, suggesting responsible observed mice. Mechanistic studies revealed acted a PPARγ target gene maintain adipocytes. Furthermore, specific knockdown fat body Drosophila led mitochondria morphology, decreased storage, made more sensitive starvation. conclusion, first time, our findings demonstrate maintaining whole-body homeostasis.

Adipocyte iron regulates adiponectin and insulin sensitivity.
Iron overload is associated with increased diabetes risk. We therefore investigated the effect of iron on adiponectin, an insulin-sensitizing adipokine that is decreased in diabetic patients. In humans, normal-range serum ferritin levels were inversely associated with adiponectin, independent of inflammation. Ferritin was increased and adiponectin was decreased in type 2 diabetic and in obese d...

Objective(s):In insulin resistance, the insulin action in liver, muscles and adipocytes decreases and result in hyperglycemia, dyslipidemia and hyperinsulinemia. In this study we evaluate the effect of Zataria multiflora extract on insulin sensitivity in high fructose fed insulin resistant rats, since this extract was shown antihyperglycemic effect in streptozotocin induced diabetes in rats. ...
متن کاملAdipocyte NCoR Knockout Decreases PPARγ Phosphorylation and Enhances PPARγ Activity and Insulin Sensitivity
Insulin resistance, tissue inflammation, and adipose tissue dysfunction are features of obesity and Type 2 diabetes. We generated adipocyte-specific Nuclear Receptor Corepressor (NCoR) knockout (AKO) mice to investigate the function of NCoR in adipocyte biology, glucose and insulin homeostasis. Despite increased obesity, glucose tolerance was improved in AKO mice, and clamp studies demonstrated...
متن کاملFarnesyl pyrophosphate regulates adipocyte functions as an endogenous PPARγ agonist
The cholesterol biosynthetic pathway produces not only sterols but also non-sterol mevalonate metabolites involved in isoprenoid synthesis. Mevalonate metabolites affect transcriptional and post-transcriptional events that in turn affect various biological processes including energy metabolism. In the present study, we examine whether mevalonate metabolites activate PPARγ (peroxisome-proliferat...
